CS-ASO nanocomplexes as non-viral gene delivery systems for treatment of Cystic Fibrosis
The group of Prof. Weber from Muenster has developed very promising therapeutic approach from nanomedicine for the disease cystic fibrosis.
K. Kolonko and colleagues address the misdirected Na+ absorption by the “epithelial sodium channel” ENaC. A nano-delivery system based on the natural chitosan (CS) transports antisense oligonucleotides (ASO) against ENaC und thus acts on Na+ hyperabsortion.
These nano-delivery systems, which are absorbed by the human respiratory epithelium, are non-viral, non-toxic and biodegradable by chitinases.
To check the successful complex binding of chitisan and the antisense oligonucleotides band-shift assays were carried out with Midori Green Advance.
