##### Long-term storage of cultured cells

Cryopreservation of mammalian cells is extremely valuable and common in biological research. Fear of losing a cell line to contamination or incubator failure is frequently the impetus for making archival storage of cells a high priority after receiving or generating a new cell line. Once transferred from growth media to freezing medium, the cells are usually frozen at a controlled rate and stored in liquid nitrogen vapor or at -130°C in a mechanical deep freeze. Although freezing a cell line is a commonly performed procedure, problems arise when suitable freezers are not available, or unwanted variables are introduced by the presence of serum, extra-wash or complicated freezing algorithms.

##### Serum adds variation to long-term storage

All Bambanker™ products are produced with no serum. Cryopreservation media which contain sera have the disadvantage of fluctuations in recovery rates and undefined composition. Reproducibility of experiments with cells which were frozen in a serum-containing-medium, could be affected by lot-to-lot variation of the serum since the composition and concentration of proteins and other biological molecules varies with each batch of serum. This may result in issues when thawing and using cells from such serum-containing media. ##### Comparison of Bambanker™ with another cryopreservation medium for the cultivation of 1,400 different cell lines

The JCRB cell bank handles approximately 1,400 different cell lines. A low survival rate after thawing frozen cell lines (KHYG-1, KAI3, HL60, OVMANA) has let them to test Bambanker™ and compare it to the up to then used preservation medium for the four cell lines. The growth efficiency after thawing was compared for cells stored with the currently used commercially available preservation medium and Bambanker™.

All cultured cells were harvested in the logarithmic growth phase. 1 ml preservation media was added to approximately 1 x 106 cells in a storage tube. The cells were stored for 2 weeks at -80°C. The frozen cells were thawed in an 37°C water bath and incubated at 37°C and 5% CO2 in a 96-well plate. Every day the viable cell number was determined.

The survival rate after thawing of the four cell lines (KHYG-1, HL60, KAI3, OVMANA) is with the currently used commercially available product and Bambanker™ very low. However, after thawing, all four cell lines cell proliferation was improved with Bambanker™ when compared to the currently used commercial product.

##### Is Bambanker™ serum-free?

Yes. Bambanker is a serum-free cryopreservation medium.

All Bambanker™ products are produced without fetal bovine serum. This helps reduce lot-to-lot variation and avoids undefined serum components in cryopreservation workflows.

##### Does Bambanker contain DMSO?

Yes. Standard Bambanker contains DMSO. If you need a DMSO-free cryopreservation medium, use Bambanker™ DMSO Free ([BBF01](https://www.nippongenetics.eu/en/products/cell-biology/freezing-media/dmso-free-media/bambanker-dmso-free/)).

Bambanker™ DMSO Free is the only Bambanker version without DMSO and is recommended for highly sensitive cells. For standard cell lines, the regular Bambanker™ formulation can be used as a ready-to-use serum-free freezing medium.

##### Does Bambanker™ contain antibiotics or antimycotics ingredients?

#### Usage

##### Can we thaw cells directly in culture medium, without a washing step?

Yes. Cells frozen with Bambanker™ can be thawed directly into culture medium without an additional washing step.

Bambanker is designed as a ready-to-use cryopreservation medium for a simple freeze-thaw workflow. Always follow the protocol recommended for your specific cell type and downstream application.

##### What cell number is recommended for freezing with Bambanker™?

**The recommended cell number is 1 × 10⁶ cells per 1 mL Bambanker™.**

A practical working range is approximately 5 × 10⁵ to 1 × 10⁷ cells per 1 mL Bambanker. Avoid substantially exceeding this range, as very high cell densities may reduce post-thaw survival.

##### What is the minimum volume of Bambanker™ that can be used?

**As a standard recommendation, use 1 mL Bambanker**™ **for 1 × 10⁶ cells. Smaller volumes can be scaled proportionally, for example 0.5 mL for 5 × 10⁵ cells.**

When reducing the volume, keep the cell-to-medium ratio consistent and validate the result for your specific cell type. For sensitive cells or critical applications, start with the standard protocol before scaling down.

##### Can Bambanker™ be used directly from the refrigerator?

**Yes. Bambanker can be used directly from refrigerated storage. It does not need to be brought to room temperature before use.**

Bambanker is supplied as a ready-to-use freezing medium. Keep handling time short and work according to your standard sterile cell culture procedure.

##### Is a controlled-rate freezer required when using Bambanker?

**No. Bambanker does not require programmed or sequential freezing.**

The medium is ready to use and designed for a simplified freezing workflow: harvest cells, remove medium, resuspend cells in Bambanker, transfer to cryovials, and store at -80 °C or in liquid nitrogen.

##### Can Bambanker be used for long-term storage at -80 °C?

**Yes. Bambanker can be used for long-term cell storage at -80 °C or in liquid nitrogen.**

##### How can organoid survival be improved when using Bambanker?

**For organoids or tissue fragments, standard Bambanker can be used, but smaller tissue pieces may improve freezing performance.**

For tissue fragments, reduce the tissue size as much as practical before freezing. We would also recommend making the tissue finer than 20 mg. A short pre-incubation in Bambanker at 4 °C for about 10 minutes before freezing may also help. #### Applications

##### Can Bambanker™ be used for Drosophila?

**Bambanker can be used for Drosophila-derived cells. Whole-organism cryopreservation is not covered by the available Bambanker experience on this product page.**

For best results, use Bambanker with isolated cells and validate the protocol for your specific Drosophila cell type or experimental model.

##### Can it be used for secondary cells.

Yes, Bambanker can be used for secondary cells.

##### Can cells frozen in Bambanker™ be used for RNA isolation?

**Yes. Cells frozen in Bambanker can be used for RNA isolation.**

However, the RNA yield depends strongly on the number of cells used. If only around 1 × 10⁶ cells are frozen, the total RNA amount may be low, depending on the cell type and RNA isolation method.

##### Can Bambanker™ be used for freezing viruses in infected cells?

**Yes. Bambanker can be used to freeze virus-infected cells, provided that the infected cells remain viable and suitable for cryopreservation.**

The key factor is the health and stability of the host cells before freezing. Optimize the timing of harvest and freezing according to the infection model and downstream assay.

    Wir verwenden das Einfriermedium Bambanker für unsere PDX-Leukämiezellen. Bisher konnten wir feststellen, dass die Zellen nach dem Auftauen etwa 20–30 % vitaler sind. Die prozentuale Steigerung der Vitalität nach dem Auftauen erscheint zwar relativ gering, stellt jedoch bei humanen Leukämiezellen einen signifikanten Unterschied dar. Aktuell frieren wir auch verschiedene Zelllinien ein, damit haben wir jedoch noch keine Erfahrung.

    We use the Bambanker cryopreservation medium for our PDX leukaemia cells. So far, we have found that the cells are approximately 20–30% more viable after thawing. Although the percentage increase in viability after thawing appears relatively small, it represents a significant difference in human leukaemia cells. We are currently also freezing various cell lines, but we do not yet have any experience with this.

    We had the opportunity to work with the Bambanker medium to preserve our L929 fibroblasts and THP-1 monocytes. This product works well, as we experienced no issues with the thawing/freezing processes. It is a nice product for those who don’t have the facilities for liquid nitrogen preservation at -196°C.

    We have been using the Bambanker for several years, and with extreme satisfaction, to freeze our monolayer cell lines. We use it for several cell lines (BHK21, VERO, MDBK, TURB BOV…) and also for other cell lines more susceptible to freezing such as for example a primary line made in our laboratory. With the Bambanker we freeze at different temperatures, below -80°C and in nitrogen, and we have experienced greater recovery in less time than our previous freezing medium. We are very satisfied and will continue to use it.

    Wir nutzen Bambanker seit einiger Zeit als Standardeinfriermedium für verschiedene Zelltypen: Tumorzelllinien, Hybridomzellen, Primärzellen (PBMCs). Es funktioniert für alle Zelltypen sehr gut und zuverlässig und vereinfacht viele Abläufe bei uns. Außerdem wird der internationale Versand enorm vereinfacht, da kein Serum enthalten ist und trotzdem, auch bei längeren Laufzeiten und Aufenthalt im Zoll, auf Trockeneis transportiert werden kann. Auch die Lagerung bei -80°C funktioniert gut und erleichtert vieles.

    We have been using Bambanker as a standard freezing medium for various cell types for some time: Tumour cell lines, hybridoma cells, primary cells (PBMCs). It works very well and reliably for all cell types and simplifies many procedures for us. It also simplifies international shipping enormously, as it does not contain any serum and can still be transported on dry ice, even with longer transit times and stays in customs. Storage at -80°C also works well and simplifies many things.

    Mit der Verwendung des Bambaker Einfriermediums konnten wir den prozentualen Anteil überlebender iPS-Zellen nach dem Auftauen wesentlich erhöhen. Wir sind mit diesem Produkt überaus zufrieden und verwenden es mittlerweile für alle Stammzellkulturen.

    Since many years we are working with primary cell cultures derived from high-grade brain tumors which are highly susceptible to environmental changes and differing culture conditions. Thus, freezing and thawing is a challenging process for these cells. According to our experiences, we could not avoid to lose 30% to 40% of living cells after each freezing/thawing cycle. In addition, the cells need a recovery time of at least five days after freezing in our previous freezing medium, obtained from other manufacturers. We’ve recently tested the Bambanker medium with striking results: recovery times of only two days of 80% of living and “healthy” cells. We observed this improvement of recovery time and proportion of living cells with all four cell lines, that we’ve tested so far. We are very satisfied with Bambanker and will continue to use it.

    Wir arbeiten seit vielen Jahren mit primären Zellkulturen, die aus weitfortgeschrittenen Gehirntumoren gewonnen werden. Diese Zellen reagieren sehr empfindlich auf Umweltveränderungen und sich ändernde Kulturbedingungen. Das Einfrieren und Auftauen wird damit zu einem schwierigen Prozess für diese Zellen. Unseren Erfahrungen nach können wir einen Verlust von 30 % bis 40 % der lebenden Zellen nach jedem Einfrieren/Auftauen Zyklus nicht vermeiden. Zusätzlich benötigen die Zellen nach dem Einfrieren in unserem vorherigen Medium eines anderen Herstellers eine Erholzeit von mindestens fünf Tagen. Kürzlich haben wir das Bambanker Medium mit faszinierenden Resultaten getestet: Erholzeiten von nur zwei Tagen von 80 % der lebenden und “gesunden” Zellen. Wir konnten diese Verbesserung der Erholzeit und die Proportion von lebenden Zellen bei allen vier Zelllinien, die wir bisher getestet haben, beobachten. Wir sind sehr zufrieden mit Bambanker und werden es weiterhin benutzen.

    JCRB cell bank has carried out cell bank business for 30 years and we currently store 1400 types of cell lines. In 2013, we offered about 4,300 ampules for a fee to domestic and foreign researchers. Due to the high number and the wide variety of cell lines, we had some problems.
    Thus some users complained that their cell lines of dying after thawing, resulting in unsuccessful cultivation. Especially four types of cell lines were a problem which had to be urgently improved. Therefore, we compared Bambanker™ with our currently used commercial preservation medium in a cryopreservation test. The cell lines, which were stored with Bambanker™, showed much higher cell proliferation than cells, which were stored with our currently used commercially available product. Surprisingly, with Bambanker™ we got for all four cell lines very reproducible results.
    In addition, Bambanker has no differences between the lots since it does not contain serum. We are very grateful for that because it strongly simplifies the delivery procedure overseas. In the future, we will completely change to Bambanker™ in order to improve the survival rate and growth of our cells. We are thankful for resolving that long-standing problem and recommend Bambanker™ to all domestic researchers and foreign cell banks.
    This data will be also described at our JCRB website. In the near future we plan to test Bambanker™ for human tissue-derived and human iPS cell lines.
