Reliable probe qPCR – from routine assays to multiplex workflows
Clara® Probe Mix is a concentrated 4x master mix for sensitive probe-based DNA detection. The optimised formulation supports reproducible amplification across both singleplex and multiplex assays while leaving additional reaction volume available for template, primers and probes.
Powered by an antibody-mediated hot start Taq DNA polymerase, Clara® Probe Mix is compatible with a broad range of probe chemistries and standard or fast-cycling real-time PCR instruments. It is suitable for routine research workflows as well as more complex high-throughput and multiplex assay development.

Lo-ROX, Hi-ROX or No-ROX?
The appropriate ROX concentration depends on the real-time PCR system used. Clara® Probe Mix is therefore available in different reference dye configurations.
- No-ROX: Without a passive ROX reference dye
- Lo-ROX: Low ROX concentration
- Hi-ROX: Hohe ROX concentration
- Separate-ROX: ROX is supplied separately and can be dosed flexibly

Sensitive detection even at low template concentrations
Clara® Probe Mix enables reproducible amplification across a broad range of template concentrations and supports reliable detection even at low template levels.
Experimental details
Amplification of common housekeeping genes (GAPDH, and β-2-Microglobulin [B2M]) using Clara® Probe Mix (pink curves) or TaqMan Fast Advanced Master Mix (grey curves). Amplification curves are shown in the left panels and efficiency in the right panels. Three (for GAPDH) or four (for B2M) serial dilutions of mouse cDNA template were used, corresponding to 1 ng/μL, 100 pg/μL, and 10 pg/μL, or 1 ng/μL, 100 pg/μL, 10 pg/μL, and 1 pg/μL, respectively. The reaction volume was 20 μL. The cycling conditions were: 95 °C 2 min, followed by 50 cycles of 95 °C 10 s, 60 °C 30 s. Clara® Probe Mix overperforms TaqMan Fast Advanced Master Mix, showing high sensitivity and reproducible amplification even at low template concentrations with optimal efficiency. Multiplex qPCR on the FastGene® qFYR Real-Time PCR System

Experimental details
Clara® Probe Mix was used in a 6-plex reaction on a FastGene® qFYR Real-Time PCR System. Two technical replicates were used for each target. All 6 targets were successfully amplified in a single well in both replicates.
