Optima HotStart Ready Mix- It is your decision when to start

The FastGene® Optima HotStart Ready Mix combines the superb efficiency and robustness of the FastGene® Optima with a proprietary antibody that inhibits preliminary unspecific reaction. This antibody is destroyed during the primary activation step.

Opima(l) Convenience – Just add template and primers

The ReadyMix version of the FastGene® Optima HotStart is the easiest way to get all of these advantages. It comes with all the necessary ingredients for an optimal PCR, just add a template and primers. Additionally, the ReadyMix comes with a loading dye, meaning that the PCR sample can be directly loaded onto an agarose gel, saving your precious time for what really matters.

  • RT-PCR
  • Very complex templates
  • GC-rich templates
  • SNP analysis
  • Multiplex PCR
  • Any standard PCR application
1. Direct PCR from Escherichia coli colonies

“With the FastGene® Optima HotStart Ready Mix with Dye we could clearly distinguish between samples with inserts (800 bp) and those without inserts (300 bp).”

Fig. 1: Direct PCR from Eschrichia coli colonies using FastGene® Taq Optima HotStart Ready Mix or Competitor T. The Ready Mixes were used to determine the presence or absence of inserts. The FastGene® Taq Optima HotStart Ready Mix yielded a clearer electrophoresis pattern without smearing and delivered a result for all colonies. The competitor T was not able to amplify 10 colonies.

2. Genotyping using mouse-tail tissue

“Analysis of the experiment was very easy because it already contains the loading dye, meaning that we could apply the reaction directly onto an agarose gel. Additionally, the PCR efficiency was higher when compared to the competitor‘s ready-mix without loading dye”

Fig. 2: Genotyping of using FastGene® Taq Optima HotStart Ready Mix or a competitor. The two Ready Mixes delivered the same genotyping results. The FastGene® Taq Optima HotStart Ready Mix had a higher efficiency shown by the bigger bands. The FastGene® Taq Optima HotStart Ready Mix is easier to use since it contains the loading dye meaning that the PCR product can be directly added to the gel.

What is the error rate of this enzyme?

The error rate is around 10-15 times lower compared to a regular TAQ. The FastGene Optima PCR system is a blend of Taq DNA polymerase and an engineered archaeal (B-family) DNA polymerase. Both enzymes possess 5’ – 3’ polymerase activity, but only Taq possesses 5’ – 3’ exonuclease activity, and only the B-family DNA polymerase possesses 3’ – 5’ exonuclease activity.

Does the PCR products have a 3′ A overhang?

Yes there is a 3′ A overhang.

Can I use Optima for dicret PCR from cell lysates without DNA extraction?

This should work for bacterial cells. For eukaryotic cells, we recommend to use DNAreleasy.

2 reviews for FastGene Optima HotStart Ready Mix

  1. Dr. Matthias Schmidt; Institute for molecular life science, Frankfurt

„Wir haben den HotStart ReadyMix für Duplex-PCR von cDNAs unserer knock-down Mutanten mit sehr gutem Erfolg getestet. Die Reaktionen zeigen kaum unspezifische Produkte und sind reproduzierbar. Außerdem hat dieses Produkt ein sehr gutes Preis/Leistungs-Verhältnis.“

    Translation by Nippon Genetics

“We tested very successfully the HotStart ReadyMix for duplex-PCR of cDNAs from our knock-down mutants. The PCR reactions show hardly unspecific products. Additionally this product has an excellent price performance ratio.”
